Obtaining DNA of good quality for PCR or sequencing is the first step in most of the current molecular biology applications. Many protocols are cumbersome to apply and use toxic products. We compared a method commonly used in many laboratories with a more simplified method ? direct colony PCR ? that does not use any hazardous reagents and equipment. We employed species belonging to the Mycosphaerella complex, and used the DNA for PCR and sequencing. The results showed that the direct colony PCR generated the expected band sizes when the primers ITS1-5.8s-ITS2, and for specific sequences for M. fijiensis or M. musicola were used. Furthermore, the sequencing of the ITS-amplified region showed a 100% similarity between the two methods. |
